E. coli strains

General
Statens Serum Institut has a strain bank containing more than 62,000 E. coli strains, of which most are clinical isolates. The collection contains both wild types and reference strains.
Product description
The reference strains are primarily sold as freeze-dried ampoules, whilst most wild type strains are sold as agar slant in transport medium. Freeze-dried strains can be stored for many years in unopened ampoules at room temperature. Once the ampoule is opened, the strain must be used immediately. Strains sent in agar slant must be plated out within a week from receipt and resown, as strains cannot survive in transport agar for long. All strains are typed for the antigen they are sold under just before sale.
Use
The strains sent in agar slant can be sown directly out onto a plate, while the freeze-dried ampoules must be added to 1 mL bouillon, incubated overnight at 37°C, and the entire ampoule contents must be transferred onto a plate. The plate is incubated overnight at 37°C, and the strain can now be used for the intended purpose, or frozen slants can be made of it. E. coli strains that cannot be typed can be sent to Statens Serum Institut, the International Escherichia Centre (WHO), Building 37K, Room 125C, Artillerivej 5, DK-2300 Copenhagen S, Denmark, for further investigation.

The strain bank also contains wild type strains representing a wide selection of the possible serotypes and virulence factors in various combinations. When ordering a wild type strain, it is important to specify which antigen/virulence combinations are required, as our strain bank, as mentioned, consists of more than 62,000 strains. For example, an order may appear as follows:
A) An O149:H18 strain. The rest of the antigens and virulence factors are unimportant.
or
B) Three strains that all contain the eae gene. Serotype irrelevant.
or
C) Six O26 strains with different H antigens.
Please note concerning the OK O serotypes, that except for the OK O strains OK O26, OK O45, OK O55, OK O114 and OK O157, the other OK O serotypes are matched with the O strains with the corresponding number e.g. OK O86 antisera is matched with the O86 strain.

Q: What is the difference between reference strains, test strains and wild type strains?
A: A reference strain is a microorganism defined to at least the genus and species level, catalogued and described according to its characteristics e.g. O, K and H serotype and stating its origin.
A test strain is generally used for quality control and performance testing of culture media or serving as a positive/negative control in a diagnostic kit ensuring that the performance of the test has been successful.
Wild type strains are isolates from patients, animals or other sources, and these are not well defined.
Q: How long time does a serotyping at SSI take?
A: It can usually be done within three weeks.
Q: How should a strain be sent?
